Aline DNA Normalizer Kit V2 leverages Aline’s proprietary magnetic bead technology featuring fixed DNA binding surfaces. Users can isolate a pre-defined amount of DNA by simply limiting the bead volume added to the reaction. This kit accomplishes DNA normalization during the purification process, eliminating the need for additional quantification and dilution. By consolidating these steps into a single process, the system reduces time, labor, and reagent costs. All DNA fragments from PCR amplification or enzymatic reactions can be normalized and combined for streamlined Next Gen library construction.
Note: The DNA output following the standard protocol is about 100ng per reaction. However, the output can be adjusted by end-users if desired.

Process Overview
The protocol mainly consists of binding, washing and elution steps, and can be performed directly in the thermal cycling plate and requires no centrifugation or filtration. The process can be automated with a walk away solution for high throughput applications.
- Bind PCR products or genomic DNA to magnetic beads, then separate beads on magnet plate.
- Wash beads with water to remove excessive DNA, nucleotides, salts, and other contaminants.
- Elute DNA into a Tris buffer.
