Product Details
The RNA Easy Kit utilizes Aline’s proprietary paramagnetic bead technology to isolate total RNA from PAXgene® preserved blood. This highly versatile system first lyses blood samples and digests proteins to ensure high-yield recovery. Subsequently, the magnetic particles immobilize the DNA/RNA, allowing for the rapid removal of contaminants.
The simple recovery procedure includes an on-bead DNase I treatment to remove DNA. Users can easily modify the protocol to isolate genomic DNA by omitting the DNase I step. This flexible system also supports specialized microRNA isolation and integrates seamlessly into automated high-throughput workflows.
The process first lyses blood samples and digests proteins. Subsequently, DNA and RNA immobilize onto the magnetic particles to allow for the removal of contaminants. A DNase I treatment removes the DNA, while a simple procedure recovers the RNA. Users may modify this protocol for microRNA and DNA isolation. Genomic DNA can be isolated if the user chooses not to perform a DNase I digestion.
Key Features
- Protocol can be performed in both 96-well plate and single tube formats
- 400 μL of preserved blood sample can be processed in a 96-well plate or in a 1.5 mL tube format, and a 24-well plate may be used to process a large volume of blood
- Process is fully automated, except for initial proteinase digestion
- No brownish or greenish color in eluate
- No plate transfer is needed
Recommended Automation Platform
- Using the Beckman Coulter RNAdvance for Paxgen preserved blood script on Biomek platform is recommended; the protocols are the same (but reagents are not interchangeable)
Protocol
RNA Easy for Paxgen blood tube-Protocol v8.2
